The rapid propagation technique of shoot tip tissue culture is an effective method for eliminating or reducing viruses in lotus roots, restoring the desirable traits of varieties, increasing the propagation rate, and accelerating the dissemination of high-quality strains. In recent years, Xiangshui Shallow Water Science and Technology Demonstration Park has implemented this advanced technology, gradually achieving industrial production of shallow water seedlings. First, initial culture begins in mid-April when the lotus roots start to sprout. The process involves selecting explants from fresh seedlings, specifically the stem tip (bud). The leaves are disinfected, and the sheaths are carefully removed using tweezers. When the stem tips reach a diameter of about 2 mm, they are inoculated into a differentiation medium. After approximately 30 days of culture, multi-leaf seedlings can be successfully differentiated. Second, proliferation and rooting culture follow. 1. Subculture Propagation: Under aseptic conditions, the primary culture seedlings are cut into explants with one spread leaf, one unexpanded leaf, and one shoot. These are then placed individually into a proliferation medium. After about 30 days, the cells are harvested and transferred to a basal culture for further multiplication. The optimal period for proliferation is 30 days. With proper conditions, four generations can be completed within 120 days, resulting in a theoretical propagation coefficient exceeding 2000 times. 2. Rooting Culture: Once a sufficient number of subcultured seedlings are obtained, they are transferred under sterile conditions to a rooting medium. After 30 to 40 days, a well-developed root system forms, allowing the plant to become a complete nursery-ready seedling. Third, domestication and transplanting take place. 1. Domestication: The rooted seedlings are moved from the lab to the field together with their bottles. They are kept at around 25°C, shaded, and kept moist. The bottles are gradually opened over a week to help the seedlings adapt to outdoor conditions. 2. Transplanting: Carefully remove the seedlings from the bottle, rinse them with water, and sterilize the soil in a shallow layer of 3 to 5 cm. Plant them with a row spacing of 1 meter and a plant spacing of about 0.5 meters. Maintain a temperature of around 25°C with appropriate shading. After about 15 days, the seedlings will grow new leaves. Following standard field management practices, after being transplanted into the open field, the seedlings need a growth period of at least 100 days with temperatures above 15°C before they can produce larger rhizomes for planting the following year.
